Job summary

A highly motivated Postdoctoral Researcher is sought to join a project focused on single-cell proteomics and mass spectrometry, specifically targeting transcription factors in human blood stem cells. This role involves developing innovative workflows to detect MiTag-derived peptides alongside global proteomes, contributing to a groundbreaking interdisciplinary project.

Qualifications

  • PhD in biochemistry, (bio)analytical chemistry, cell biology, or a related field.
  • Extensive hands-on experience with LC-MS/MS-based proteomics, with at least 2–3 years of operation of high-end instruments (preferably Orbitrap series).
  • Strong background in DIA-based proteomics workflows, including hybrid targeted/untargeted acquisition strategies.
  • Demonstrated experience in single-cell proteomics, including low-input sample preparation and liquid handling automation.
  • Proficiency in computational proteomics data analysis and downstream analysis in R or Python.
  • Proven ability to conduct independent research from conception to publication.
  • Excellent written and oral communication skills in English.

Responsibilities

  • Design, optimize, and deploy scp-MS workflows for detecting MiTag peptides and global proteomes in primary human hematopoietic cells.
  • Develop DIA-based scp-MS acquisition methods on the Orbitrap Astral.
  • Optimize protocols for intracellular MiTag staining compatible with high-sensitivity scp-MS.
  • Benchmark MiTag performance against orthogonal assays.
  • Collaborate with other labs to refine reagents and transfer workflows to primary samples.
  • Publish results in leading journals and present at international conferences.
  • Mentor junior lab members and contribute to a collaborative lab culture.

Skills

  • Experience with targeted proteomics approaches and FACS-based cell sorting is desirable.
  • Familiarity with mammalian cell culture, particularly primary hematopoietic cells.
  • Ability to analyze and interpret larger single-cell datasets.

Education

  • A formal qualification of a PhD degree (or equivalent) is required.

Tools

  • Experience with DIA-NN, MSFragger, Spectronaut, or similar tools for data analysis.
